چکیده

Title of Thesis: Characterization of the Interaction of the Terminal Complement Complexes with a Model Membrane System: Comparison of the Channel Properties of CS-8 and CS-9 Lesions Rikki Solow, Doctor of Philosophy, 1987 Thesis directed by: Valerie w. Hu, Ph.D. Department of Biochemistry The interaction of CS-8 and CS-9 complexes with model membranes was investigated using large unilamellar vesicles (LUV's) prepared by reverse phase evaporation. In particular, CS-8 and CS-9 channel activity with regard to channel pore size, mode of marker release and dynamics of channel formation, was characterized. This was achieved by measuring the release of entrapped fluorescent dye 6-carboxyfluorescein (6CF), and/or radiolabeled markers, from complement-treated LUV's. CS-8and CS-9-induced permeability changes and the respective pore sizes of the complexes in LUV membranes were similar to those induced by complement in na~ural membranes, suggesting that the lesions formed on these model membranes by CS-8 and CS-9

A Survey of Anticancer Effect of Some Five- and Six-Membered Palladium (II) Complexes (Containing Bidentate Phosphorus Ligands) on Caco-2 Colon Cancer Cell Line

Background and Objectives: Transition metal complexes possess a variety of biological and pharmacological activities, such as antitumor, antimicrobial, anti-inflammatory, anti-diabetic, and free radical scavenging. Many studies have been carried out on platinum-based chemotherapy agents, however, most attention has been paid to non-platinum metal-based agents in order to find different metal comp...

Monoclonal antibodies against neoantigens of the terminal C5b-9 complex of human complement.

Assembly of the terminal C5b-C9 complement components into the cytolytic C5b-9 complex is accompanied by exposure of characteristic neoantigens on the macromolecule. We report the production and characterization of mouse monoclonal antibodies to C9-dependent neoantigens of human C5b-9. Binding-inhibition assays with EDTA-human plasma and micro-ELISA assays with purified C9 showed that the antib...

Production of Ultrastructural Membrane Lesions by the Fifth Component of Complement

A direct quantitative relationship has been demonstrated between the number of cell bound C4,2 complexes or C5 molecules and the number of ultrastructural lesions visualized on the cell membrane subsequent to immune hemolysis. When bound C4,2 complexes exceeded bound C5 molecules, the number of ultrastructural lesions seen corresponded to the number of C5 molecules. However, in the reverse situ...

Expression and characterization of the C345C/NTR domains of complement components C3 and C5.

Complement components C3, C4, and C5 are members of the thioester-containing alpha-macroglobulin protein superfamily. Within this superfamily, a unique feature of the complement proteins is a 150-residue-long C-terminal extension of their alpha-subunits that harbors three internal disulfide bonds. Previous reports have suggested that this is an independent structural module, homologous to modul...

Thrombin generates previously unidentified C5 products that support the terminal complement activation pathway.

The coagulation and complement pathways simultaneously promote homeostasis in response to injury but cause tissue damage when unregulated. Mechanisms by which they cooperate are poorly understood. To delineate their interactions, we studied the effects of thrombin and C5 convertase on C5 in purified and plasma-based systems, measuring release of the anaphylatoxin C5a, and generation of C5b, the...
